Instructions for Proper Installation of an Anaerobic Incubator

Close and seal the doors of the sampling room, both inside and out, and perform a vacuum check.

2. Connect and inspect the air line, and seal the joints with sealant as necessary to prevent leaks; To form an anaerobic environment in the operation room: place the necessary accessories and equipment according to the usage requirements, and put two non-toxic plastic bags into the operation room; Turn on the power to switch on the lighting, turn on the temperature controller, and adjust the required temperature and safety temperature;

3. Turn on the UV sterilization lamp for indoor sterilization, with the sterilization time based on specific experiments.

4. The anaerobic incubator should be placed in a location with minimal temperature variation and easy to operate, avoiding direct sunlight and keeping a distance from heating equipment, and should be placed flatly; securely position the mixed gas cylinder and nitrogen cylinder flatly, and install the pressure regulators (including pressure gauges) separately, and place them in an appropriate location.

5. Operation Room Nitrogen Displacement: A. Insert a rubber hose into the operation room's intake port, and the other end into a plastic bag; B. Connect the nitrogen intake line, open the nitrogen control valve, fill both plastic bags with nitrogen, close valve 1, and then tie the bag shut; C. Place the latex gloves over the observation panel flange and secure them; D. Gradually release the nitrogen from the plastic bag into the operation room until all is dispersed.

6. Open the oxygen scavenger in the operating room, connect the power to the oxygen scavenging catalyst, and perform catalytic oxygen removal. After one hour, open the Methylene Blue indicator and observe its color change; no color change indicates that the operating room has reached an anaerobic environment.

7. Operation Room Third Gas Exchange (Mixed Gas Exchange): A. Open the gas circuit and turn on the mixed gas valve to allow air intake. During inflation, first evacuate the sampling chamber to a vacuum state and then use the foot switch to control exhaust as needed; B. After the mixed gas fills the plastic bag, close the mixed gas valve and open the valve to allow the mixed gas to pass through the flowmeter. Adjust the flowmeter to maintain a flow rate of approximately 10ml per minute; C. Gradually release the mixed gas inside the plastic bag into the operation room; D. After three gas exchanges, the oxygen content in the operation room is at an extremely low level.

8. The control room D performs a second nitrogen displacement (nitrogen replacement) by repeating the nitrogen filling process. The sampling room is first evacuated, and attention is given to using the foot switch to open and close the exhaust at all times.
